Mammalian Cell Gene Mutation Test

The in vitro mammalian cell gene mutation test can be used to detect gene mutations induced by chemical substances.  The test measures mutation to hypoxanthine-guanine phosphoribosyl transferase (HPRT) gene. Cells in cultures are exposed to four concentrations of the test substance, with and without metabolic activation. In selective medium, after mutation cells proliferate, whereas non-mutated cells cannot. Viable cells form colonies. These colonies are stained, and the mutation rate is expressed as the number of mutant colonies resulting from test article exposure. The mutation rate is compared with the cytotoxicity of the test material.
