Overview

OptiSafe™ is a shelf-stable, nonanimal test method designed to determine whether a chemical or mixture is an ocular irritant, without using animal tissues, cells, or live animals of any kind. This abstract, presented at the Society of Toxicology Annual Meeting, describes the latest round of protocol optimizations that improve the test's accuracy across a broader range of chemical classes. The work focuses on reducing the remaining sources of misclassification and expanding the validated chemical space, moving OptiSafe™ closer to full regulatory acceptance as a standalone replacement for the Draize rabbit eye test.

Abstract

OptiSafe™ is a shelf-stable, nonanimal test method to determine whether a chemical or mixture of chemicals is an ocular irritant. The test uses a macromolecular substrate to detect the irritation potential of test substances through a colorimetric endpoint. Previous work established the core protocol and demonstrated strong performance for aqueous formulations and surfactant-containing mixtures. This abstract presents further optimization work aimed at improving performance for additional chemical classes, including organic solvents, solid materials, and complex mixtures. Protocol modifications were evaluated against an expanded reference chemical set with established GHS and EPA classifications. The optimizations resulted in improved sensitivity and specificity relative to the previous protocol version, with particular gains for chemical classes that had previously shown borderline performance. The updated protocol maintains the shelf-stable, cell-free format that makes OptiSafe™ accessible to standard analytical laboratories without specialized cell culture infrastructure.
